What is 7,8-DHF?

7 8 DHF, or 7,8-dihydroxyflavone, is a flavonoids compound, exist in nature but rare to see. 7,8-DHF can mimic the function of brain-derived neurotrophic factor (BDNF) to act as the specific small molecular tropomyosin-related kinase B (TrkB) agonist, which can trigger TrkB dimerization to induce the downstream signaling.

In order to know the 7,8-DHF, we should know the TrkB and BDNF firstly. TrkB (Tyrosine Kinase receptor B) is encoded by the TRK family of proto-oncogenes. Three tyrosine kinase receptors (TRKs) have been cloned, namely TrkA, TrkB and TrkC. It is a transmembrane protein composed of extracellular glycosylated polypeptide, transmembrane domain and intracytoplasmic tyrosine kinase domain located on nerve cell membrane. It is a specific receptor for brain-derived neurotrophic factor (BDNF). BDNF is the most abundant neurotrophic factor in the body, and it works by combining with TrkB. It is a potential therapeutic target in numerous neurological, mental and metabolic disorders.

However, brain has a poor absorption of BDNF, yet scientists haven’t found a efficient method to deliver BDNF into body. While the scientists was researching the BDNF, 7,8-DHF was found. 7,8-DHF not only has similar effects with BDNF, but also can cross the brain barrier and get a better absorption.
