What is TEE?

Tianeptine ethyl ester, chemical name is 7-[(3-chloro-6,11-dihydro-6-methyl-5,5-dioxidodibenzo[c,f][1,2] thiazepin-11-yl)amino ] heptanic ethyl sulfate, also known as tianeptine-001, or tianeptine EP impurity B, often abbreviated as TEE, is a derivative of tianeptine. Tianeptine is a drug mainly used to treat the major depressive disorder. From the point of structure, it is classified as a tricyclic antidepressant (TCA). According to clinical trials of tianeptine, it has been shown to be as effective as other more popular antidepressants such as fluoxetine and amitriptyline. However, tianeptine exhibits fewer side effects and complications than conventional antidepressants. Due to its sound effects, TEE has won a place as an antidepressant drug on the market. As a TEE manufacturer, Xi'an Songwu can supply high-quality TEE powder at a competitive price. If you are interested in this, please feel free to contact Xi'an Sonwu.

What is TEE Used for?

Tianeptine ethyl ester is classified as an antidepressant drug, as the treatment for people who suffer from depression. Depression is one of the most common mental diseases in today's society, and its primary clinical manifestations are low mood, slow thinking, decreased volitional activity, cognitive impairment, and somatic symptoms. It affects people's mental state psychologically and thus has a significant adverse effect on people's lives. Therefore, in recent years, more and more people have begun to pay attention to mental health and depression. As a result, many drugs for depression have appeared on the market.

Many factors affect depression, and the most common are the following.

1. Dopamine

Dopamine is a precursor to norepinephrine, a key neurotransmitter in the hypothalamus and pituitary glands, and the concentration of dopamine in the central nervous system is influenced by psychiatric factors.

2. Serotonin (Serum)

The main ingredient is 5-hydroxytryptamine (5-HT), a mixture of substances that nerve cells use to transmit information to each other.

3. Norepinephrine (NA)

It is a neurotransmitter synthesized and secreted by sympathetic postganglionic neurons and adrenergic nerve endings in the brain. And it's the main transmitter released by the latter; it is also a hormone, which is synthesized and secreted by the adrenal medulla, but the content is relatively low.

How Does TEE Affect the Brain?

The basic basis of antidepressant drug treatment is: clinical depression patients often show that the concentration of serotonin and norepinephrine (hormone that can maintain a high level of mood referred to as happy hormone) in the brain environment is too low, making its vitality value greatly impaired.

When we are active, neurons (nerve cells) transmit information to each other. Hormones are substances that are released when neurons transmit information to each other to activate the next neuron. Neuron A releases the quantitative hormone, neuron B withdraws the quantitative hormone, and finally completes the transmission of information. There will be a specific concentration of hormones in our brain environment; among them, the right concentration of the happiness hormone will keep us happy. And when there is release, there is consumption. Under normal circumstances, the brain maintains the concentration of hormones free in the brain environment in its way, allowing some hormones to disappear naturally. In the first case, the brain releases a substance called monoamine oxidase, which binds to various hormones and inactivates them. In the second case, the hormone-releasing neuron A, reabsorbs (or uptakes) the free hormone.

However, the depressive factor in depression will deplete the happiness hormone in the brain environment, interfere with its natural replacement process, and reduce the individual's sense of happiness; that is, the vitality value is impaired. Therefore, the basic principle of antidepressant treatment is to intervene in the natural disappearance of the happy hormone and ensure its concentration in the brain.

There are two main mechanisms of action of common antidepressants.

The first is inhibition of the reabsorption process, which blocks the reabsorption of pleasure hormones (5-HT and NA), allowing them to stay in the synaptic cleft for longer and greater quantities. This led to the emergence of two common antidepressants, SSRIs (selective serotonin reuptake inhibitors) and SNRIs (serotonin and norepinephrine reuptake inhibitors). They are not only effective against depression but also against other psychiatric disorders that often accompany depression, as well as TCAs (heterocyclic antidepressants), including tricyclic and tetracyclic antidepressants. They were once the mainstay of treatment for depression.

The second is reducing the pleasure hormones' natural depletion (5-HT and NA). By inhibiting the production of monoamine oxidase that consumes happy hormones, the purpose is to ensure the concentration of happy hormones in the brain environment. A class of antidepressants derived from it is MAOIs (monoamine oxidase inhibitors), which are very effective but rarely used as the drug of choice, mainly when other antidepressants have failed.

Although tianeptine is classified as a tricyclic antidepressant (TCA), its pharmacology differs from typical antidepressants. This is because it is thought to not act immediately by modulating monoaminergic neurotransmitters such as serotonin, dopamine, or norepinephrine. Instead, it theoretically acts on glutamatergic and glutamatergic mechanisms, causing the brain to adapt to stress and depression more easily.

Tianeptine challenges the monoaminergic hypothesis of depression and the widely supported monoaminergic mechanism that explains the effects of most known antidepressants. The current study shows that tianeptine exerts antidepressant effects by modulating the activity of glutamate receptors (such as AMPA receptors and NMDA receptors) and affects the release of brain-derived neurotrophic factor (BDNF), thereby affecting neurological plasticity. Recent research supports the role for tianeptine in regulating glutaminergic activity in the amygdala, an emotional region of the brain associated with memory. This makes it a better option for people with depression.
