What Is The Drug Diclazuril Used For
Diclazuril powder is a synthetic triazine compound used as a feed additive and veterinary drug to control coccidiosis. Other substances in this group are clazuril and toltrazuril. The chemical structure of diclazuril is 2Magne6-dichloro-α-(4-chlorophenyl)-4-(4magenta5-dihydro-3-dioxo-1-pyrrolidine-2-triazine-2 (3H)-yl) phenylacetonitrile. In chickens and turkeys infected with single or mixed strains of Eimeria, diclazuril showed anti-coccidial activity at all test doses (diclazuril acts on the asexual or sexual stage of coccidia, blocking the excretion of oocysts). In birds, the highest plasma concentration of radioactivity was observed 6 hours after a single administration. Until 72 hours after administration, the plasma radioactivity was almost entirely attributed to the unchanged diclazuril. Diclazuril cannot be used to lay eggs for human consumption.

Diclazuril Uses
Anti-coccidial drugs. Diclazuril is a triazine antiprotozoal drug that can be effectively treated by Isospora spp., Toxoplasma gondii, and Eimeria spp. Caused by the infection. And has been used to treat coccidiosis. It is also used in horses to treat equine protozoal encephalomyelitis (EPM) caused by Sarcocystis.
Toltrazuril sulfone (ponazuril) is an active metabolite found in treated horses' serum and cerebrospinal fluid (CSF). Although the exact mechanism of action is being studied, diclazuril plays its anti-S effect. Neuronal effects are produced by inhibiting the production of merozoites.
Pharmacokinetics has been studied in horses. The oral absorption rate was 1.56%, and the half-life was 43-65 hours. Other studies on horses showed that at the dose of 1 mg/kg and 0.5 mg/kg, the half-lives were 55 and 87 hours, respectively, and the peak concentrations were 0.185 and 0.1 mcg/mL, respectively. With repeated administration to a steady state, the half-lives of 0.5 and 1.0 mg/kg were 71 and 54 hours, respectively, and the peak concentrations were 0.97 and 0.9 mcg/mL, respectively. Therefore, for long-term administration, the pharmacokinetic characteristics were similar to those of 1.0 mg/kg and 0.5 mg/kg in horses.

What Is The Mechanism of Action Of Diclazuril
Understanding the complex life cycle of Eimeria is helpful to understand the site of action of anti-coccidial drugs. The life cycle consists of three consecutive stages, namely, partial reproduction, gametogenesis and spore reproduction, the first two occurring in the intestines of birds and the third in the environment.
The stage of infectious transmission is an oocyst, which contains four sporangia when forming spores, and each sporangium contains two sporozoites. After ingesting sporulated oocysts, sporozoites are released from sporangia and penetrate the epithelial cells of the intestine. This is followed by the mitotic phase of proliferation, which leads to a further stage of movement, the release of merozoites, invading other epithelial cells. This process is repeated several times, followed by gametogenesis, the sexual phase of the life cycle, which leads to the formation of new oocysts and excretion with feces. The third stage of the life cycle (sporogenesis or sporulation) occurs in the environment of the poultry house, in which spores and sporozoites are formed.
A key feature of the life cycle of Eimeria is the haploid state of the genome, which affects the selection of drug-resistant mutants. As all stages of the post-meiotic stage of the life cycle (from sporoplasmic formation to gametes), it is haploid, so it is subject to the pressure of drug selection.
